Induction of human airway epithelial to mesenchymal transition upon rhinovirus infection

چکیده
Background Structural changes of the airway, collectively referred to as airway remodeling are believed to be the underlying cause of the airway hyperresponsiveness that is characteristic of asthma. Airway remodeling is characterized by thickening of the subepithelial membrane, goblet cell hyperplasia, angiogenesis, increased smooth muscle mass, and epithelial fragility. Thickening of the subepithelial membrane, due to increased deposition of matrix proteins by fibroblasts/myofibroblasts, has been observed in children even prior to the formal diagnosis of asthma. Recent data have shown that young children who experience episodes of human rhinovirus (HRV)-induced wheezing in early childhood are at increased risk of subsequently developing asthma. The primary site of infection of HRV is the airway epithelium. Recent evidence suggests that the molecular reprogramming of epithelial cells through a process called epithelial to mesenchymal transition (EMT) may contribute to increases in fibroblast/myofibroblast in the asthmatic airway. Therefore, we hypothesize that HRV infection plays a role in early airway remodeling by triggering EMT to produce fibroblasts/myofibroblasts that cause thickening of the subepithelial membrane by depositing matrix proteins.

Role of human rhinovirus in triggering human airway epithelial-mesenchymal transition
BACKGROUND Structural changes in the airways, collectively referred to as airway remodeling, are a characteristic feature of asthma, and are now known to begin in early life. Human rhinovirus (HRV)-induced wheezing illnesses during early life are a potential inciting stimulus for remodeling. Increased deposition of matrix proteins causes thickening of the lamina reticularis, which is a well-rec...
متن کاملHuman airway epithelial cells produce IP-10 (CXCL10) in vitro and in vivo upon rhinovirus infection.
Human rhinovirus (HRV) infections trigger exacerbations of asthma and chronic obstructive pulmonary disease (COPD) and are associated with lymphocytic infiltration of the airways. We demonstrate that infection of primary cultures of human airway epithelial cells, or of the BEAS-2B human bronchial epithelial cell line, with human rhinovirus type 16 (HRV-16) induces expression of CXCL10 [IFN-gamm...
متن کاملRole of NF-kappa B in cytokine production induced from human airway epithelial cells by rhinovirus infection.
Infection of human epithelial cells with human rhinovirus (HRV)-16 induces rapid production of several proinflammatory cytokines, including IL-8, IL-6, and GM-CSF. We evaluated the role of NF-kappaB in HRV-16-induced IL-8 and IL-6 production by EMSA using oligonucleotides corresponding to the binding sites for NF-kappaB in the IL-6 and IL-8 gene promoters. Consistent with the rapid induction of...
متن کاملIL-1α mediates cellular cross-talk in the airway epithelial mesenchymal trophic unit
The bronchial epithelium and underlying fibroblasts form an epithelial mesenchymal trophic unit (EMTU) which controls the airway microenvironment. We hypothesized that cell-cell communication within the EMTU propagates and amplifies the innate immune response to respiratory viral infections. EMTU co-culture models incorporating polarized (16HBE14o-) or differentiated primary human bronchial epi...
متن کاملCrosstalk between Tumor Cells and Immune System Leads to Epithelial-Mesenchymal Transition Induction and Breast Cancer Progression
Herein, we review the current findings of how a variety of accessory cells could participate in shaping the tumor microenvironment and supporting the mechanisms by which cancer cells undertake the epithelial-mesenchymal transition (EMT). EMT, a complex of phenotypic changes, promotes cancer cell invasion and creates resistance to chemotherapies. Among the accessory cells present in the EMT, imm...
